Dopamine D1 receptor‐expressing neurons activity is essential for locomotor and sensitizing effects of a single injection of cocaine

چکیده

Dopamine D1 receptors play an important role in the effects of cocaine. Here, we investigated neurons which express these (D1-neurons) acute locomotor cocaine and sensitization observed after a second injection this drug, using previously established two-injection protocol sensitization. We inhibited D1-neurons double transgenic mice conditionally expressing inhibitory Gi-coupled designer receptor exclusively activated by drugs (Gi-DREADD) D1-neurons. Chemogenetic inhibition low dose clozapine (0.1 mg/kg) decreased cocaine-induced expression Fos striatal neurons. It diminished basal activity hyper-locomotion induced (20 mg/kg). Clozapine 0.1 mg/kg had no effect itself did not alter wild-type mice. Inhibition during first administration prevented response to 10 days later. On Day 11, stimulation Gi-DREADD blocked locomotion including sensitized mice, whereas on 12, absence inhibition, all displayed These results show that chemogenetic decreases spontaneous activity. prevents induction blocks but does reverse Our study further supports central mediating its

CREB activity in dopamine D1 receptor expressing neurons regulates cocaine-induced behavioral effects

It is suggested that striatal cAMP responsive element binding protein (CREB) regulates sensitivity to psychostimulants. To test the cell-specificity of this hypothesis we examined the effects of a dominant-negative CREB protein variant expressed in dopamine receptor D1 (D1R) neurons on cocaine-induced behaviors. A transgenic mouse strain was generated by pronuclear injection of a BAC-derived tr...

Blocking impact of clozapine on cocaine locomotor and sensitizing effects in rats.

In the present study, we attempted to determine the effects of an atypical antipsychotic drug clozapine on the locomotor activation as well as sensitization to cocaine in male Wistar rats. When given acutely to rats, cocaine (10 mg/kg, ip) increased 4-fold the animals' locomotor activity. Repeated administration (1-5 days) of cocaine (10 mg/kg, ip) to rats significantly enhanced on day 10 the l...

simulation and experimental studies for prediction mineral scale formation in oil field during mixing of injection and formation water

abstract: mineral scaling in oil and gas production equipment is one of the most important problem that occurs while water injection and it has been recognized to be a major operational problem. the incompatibility between injected and formation waters may result in inorganic scale precipitation in the equipment and reservoir and then reduction of oil production rate and water injection rate. ...
